1 Principle and application

 

Amantadine test card is used for rapid detection of amantadine residues in poultry eggs. This product is a qualitative test.
This reagent uses the specific reaction of antigen and antibody, lateral chromatography and colloidal gold technology for rapid qualitative detection of target in samples. The conjugated antigen and sheep anti-mouse IgG antibody were pre-coated on the NC film, and the drug residue in the sample and the conjugated antigen pre-coated on the NC film competed with the target antibody to inhibit the binding of the antigen and the target antibody. Therefore, by observing the color intensity ratio of the detection line and the quality control line, it can be judged whether the drug residue of the target substance in the sample exceeds the specified detection limit value.

 

2 Technical Specifications
2.1 Detection limit of poultry eggs: 2ppb (ng/ml)

 

Preprocessing of poultry egg samples

 

Notice before sample processing:
The experimental apparatus must be clean and disposable suction heads used to avoid pollution interfering with the experimental results, and the temperature of the experimental environment must be above 20 degrees.
4.1 Fresh or cryopreserved poultry eggs are shelled and homogenized, and about 4g egg pulp samples are weighed and added into 15ml centrifuge tube.
4.2 Accurately add 5ml reagent A (acidified acetonitrile) into the centrifuge tube and shake violently for 1min; Add 1ml of reagent B and 1 tube of reagent C successively, seal the bottle stopper tightly, shake violently and mix well for 2 minutes, and centrifuge at 4000rpm for 3 minutes.
4.3 Remove 3ml of supernatant into 5ml centrifuge tube and dry with nitrogen or hair dryer;
4.4 Add 1ml solubilizer to the blowdry centrifuge tube, first dissolve the residue of the tube wall completely by oscillation, then add 0.3ml AM sample resolution, fully mix for 30s, and then stand for 1 minute until obvious stratification;
4.5 Absorb the lower layer solution after 120µl stratification, which is the liquid to be tested;
