              The AFB1 /DON dual fluorescence quantitative test card applies the principle of competitive suppression immunochromatography. AFB1 , DON in the sample binds to the specific monoclonal antibody labeled with fluorescent microspheres during the flow process, and then inhibits the binding of the antibody to AFB1 , DON conjugations on the NC membrane detection line. The fluorescence signal is generated after excitation at a specific wavelength, and the intensity of the signal is inversely proportional to the AFB1 , DON contents in the sample.

This product is suitable for the quantitative detection of AFB1 , DON in grain, feedstuff and some finished feed.

• Sample extract: 50% absolute ethanol-pure water solution (volume ratio)

Procedures

1. Preparation of instruments

(1) Turn on the portable fluorescence analyzer and preheat for 5 min.

(2) Turn on the incubator and heat to 37 °C.

2. Preparation of reagent

(1) Please read the Instructions for Use carefully before the experiment, and recover the test reagents and the samples to room temperature (20-25°C) before use.

(2) Insert the data card into the corresponding port of the analyzer.

(3) Tear the seal of the aluminum foil bag and remove the test card from the aluminum foil bag. Please use as soon as possible within 30 min. At most 8 samples can be tested at one time; if there are more than 8 samples, the samples can be tested in batches by 8 samples in one group.

(4) Transfer 150 µL of sample to be tested using a pipette with an appropriate range into the microsphere freezer-dried product, purge 2-3 times, immediately take 75 µL of the above solution into the injection well of the test card, and allow to react at 37°C for 7 min (pay attention to avoid obvious bubbles when pipetting and adding samples).

(5) Insert the test card into the test slot (handle facing outward, injection well facing inward, viewing window facing upward), and press the "Quick Detection" key so that the detector will automatically detect the test card (please strictly control the reaction time of 7 min, and read the result within 1 min).

Precautions

1. Do not touch the color display area of the test strip when carrying out the test.

2. The test strip is for single only, and should not be reused. 3. If the test result is out of the upper limit of the curve range and the specific value needs to be tested, the centrifugal supernatant obtained in the second step in [Sample Pretreatment] is diluted with the sample extract (50% ethanol solution), and then the sample diluent is added, mixed well and tested according to the normal steps. The final test result should be multiplied by dilution factor of 2. 4. This method is only used for preliminary screening. If the sample is positive (sample result > 80% positive limit can be judged as suspected positive), please use the confirmation method for confirmation. 5.
